Over expression of the EGF recptor can lead to accelerated dimerisation, leading to an increased proliferative signal within the cell. Autophosphorykation can occur intracellularly, so always switched on to trigger proliferation. Truncated forms of the EGF recpter, lacks the HER2 domain, which means a poorer prognosis for the patient and lower response to trastuzumab in treatment
